Semax is a synthetic heptapeptide derived from the ACTH 4-7 fragment (Met-Glu-His-Phe), extended with a Pro-Gly-Pro sequence for metabolic stability. Developed at the Institute of Molecular Genetics of the Russian Academy of Sciences, it is approved in Russia and Ukraine as a nootropic and neuroprotective agent. Unlike standard ACTH analogs, semax lacks hormonal activity (no cortisol stimulation) but retains and enhances the peptide's neurotrophic and cognitive-enhancing properties. It significantly upregulates BDNF and activates serotonergic, dopaminergic, and cholinergic systems. Russian clinical use includes stroke rehabilitation, traumatic brain injury, and cognitive enhancement.
