GHRP-6 was one of the first synthetic GH secretagogues developed, originally synthesized in the 1980s. It produces potent GH release through GHS-R1a agonism but is distinguished from other GHRPs by its marked stimulation of appetite and hunger — an effect mediated through ghrelin receptor activation in the hypothalamus and vagus nerve. This hunger-stimulating property has made it popular in bodybuilding research for mass-gaining protocols, while limiting its appeal for body composition-focused use. Like other GHRPs, it elevates cortisol and prolactin at research doses.
